First off you have to make sure that you include iTunes tags in your podcast. There is a tutorial that will walk you through setting up an iTunes compatible podcast - http://www.feedforall.com/itunes-tutorials.htm
Once the feed is properly created, then you can login to iTunes and submit the feed to iTunes here.
